EnzymeimmunoassayforthequalitativeandquantitativedeterminationofIgGantibodiesagainstU1-RNPinhumanserum.Uracil-rich,smallnuclearRNAs(U-snRNA)arephysiologicallycomplexedwithproteinstoribonucleoproteinparticles(U-snRNP)whichareinvolvedinthesplicingprocessofpre-mRNA.Differentcomplexes(U1-U6)canbedistinguishedaccordingtotheparticipatingRNA;theirproteincomponentsareinpartcommon,inpartdifferent.Forexample,U1-snRNAiscomplexedwiththeso-calledSm-proteinswhicharealsopresentinotherU-snRNPs,andwiththeU1-specificproteinsA,Cand68kDa.AutoantibodiesdirectedagainstthesethreeU1-specificproteinsareconsideredasdiagnosticMarkerformixedconnectivetissuedisease(MCTD).Theyarealsoassociatedwithsystemiclupuserythematosus(SLE)butoccurwithdistinctlylowerprevalence(about35%)andmostlyincombinationwithfurtherantinuclearantibodies(ANA).Nowadays,proteinsA,Cand68kDaareregardedastheactualRNPantigen,ratherthanthecompleteU1-RNP.
