Rib-P-AbELISAisasolidphaseenzymeimmunoassayemployingnativehumanribosomalP-proteinsP0,P1andP2isolatedfromeukaryoticcellineforthequantitativedetectionofantibodiesagainstribosomalP-proteins(rib-P)inhumanserum.Thespecificityofanti-rib-Pantibodiesisrestrictedtoacommonantigenicdeterminantlocatedonthehighlyconservedcarboxyl-terminalportionofthethreePproteins.Theassayisatoolinthediagnosisofsystemiclupuserythematosus(SLE).TheribosomalphosphoproteinsP0(~38kDa),P1(~19kDa)andP2(~17kDa)arelocatedwithinthe60Ssubunitofhumanribosomes.Incontrasttothemajorityofbasicribosomalproteins,P1andP2areacidic.TheribosomalproteinsareassociatedtoapentamerwithtwoP1/P2heterodimersanchoredtoP0bytheaminoterminalportionofP2.ThispentamerislocatedinahighlyaccessIBLeregiononthestalkoftheribosome.BiochemicalstudiessuggestthatP1/P2playafundamentalroleinallthreephasesofribosomalpolypeptidsynthesis(initiation,translocation,termination).AutoantibodiestoribosomalproteinsarehighlyspecificforSLEsincetheyarenotfoundinotherautoimmunediseasesorininfections.Thefrequencyofanti-rib-Pantibodiesis10-20%inrandomlyselectedSLEpatients.Anti-rib-Pantibodiesaredetectedmorefrequentlyinlupuspatientswithseverepsychiatricmanifestations.Inaddition,otherorganinvolvementincludingrenalandhepaticdiseasemightbecorrelatedwiththepresenceofanti-rib-P.
