Mitochondrial(AMA)-M2-AbELISAisasolidphaseenzymeimmunoassayemployingnativemitochondialM2antigenforthequantitativedetectionofantibodiesagainstM2inhumanserum.Theassayisatoolinthediagnosisoftheprimarybiliarycirrhosis(PBC).Primarybiliarycirrhosis(PBC)isachronicinflammatorydisorderofthesmallandmediumbileductsandSEROlogicallycharacterizedbythepresenceofcirculatingM2autoantibodies.Anti-M2autoantibodiesbelongtothegroupofanti-mitochondrialantibodies(AMA).TheheterogeneouslyreactingspecificAMAoftheM2subtypearedirectedagainstthreerelatedproteinsofthea-ketoaciddehydrogenasecomplex,whichislocatedattheinnermitochondrialmembrane.ThemajorepitoperecognizedislocatedontheE2subunitandtheproteinXofthepyruvatedehydrogenasecomplex(PDC).Additionally,AMA-M2autoantibodiesrecognizethe(E1aandE1ß)subunitsofthesamecomplexandtheE2subunitofseveralothermultienzymecomplexes,suchasthe2-oxo-glutaratedehydrogenasecomplex(OGDC)andthebranchedchain2-oxoaciddehydrogenasecomplex(BCOADC).ThedeterminationofAMA-M2isapowerfultoolindiagnosingPBC.
