Interleukin-1(IL-1),originallydescribedin1972aslymphocyteactivatingfactor(LAF)foritseffectsonthymocytes,isapolypeptidecytokinewithtwomolecularforms.BothformsappeartomediateidenticalrangesofBIOLOGicalactivitywhichincludesynthesisoftheacutephaseproteinsbyhepatocytes,chemotaxisofpolymorphonucleocytes,andreleaseofpolymorphonucleocytesfrombloodandbonemarrow.EarlyresearchersalsocalledIL-1betaendogenouspyrogen,andithasbeenshowntoinducefeverandisthoughttocontributetowastingofmuscles(PIF,proteolysisinducingfactor).ThetwodistinctmolecularformsofIL-1arethoughttobederivedfromtwogenes.Aftertranscription,as31kDprecursorpolypeptideiscleavedtogiverisetomostlycellmembraneassociatedIL-1alphaandsecretedIL-1beta.Bothhavethesamemolecularweightof15kDbuthavedifferentisoelectricpointsof5and7,respectively.LowlevelsofIL-1bhavebeenreportedinnormalserum.ItisthoughtthatIL-1genesareinducedtorespondtotissuedamageorininfection.ElevatedlevelshavebeenreportedinanumberofinfectiousdiseaseconditionsandinnoninfectiousinflammatoryconditionssuchasCrohn’sdisease.Inadditiontoelevatedserumlevels,IL-1hasbeenfoundinsynovialfluidsofpatientswithrheumatoidarthritisandincerebrospinalfluidafterneurologicalinflammationorinsult.Attheotherendofthespectrum,lowlevelsofIL-1havebeenfoundinmalnutritionandadvancedneoplasiasuggestingperhapsacompleximmunologicalandphysiologicalregulatoryroleforthiscytokine.
