Interleukins17F(IL-17F)and17A(IL-17A)arecloselyrelatedmembersoftheIL-17cytokinefamily,sharingabout50%aminoacididentity.StudiesinthemousehaveidentifiedTh17cellsasadistinctCD4+TcelllineagethatisdefinedbytheproductionofIL-17FandIL-17A.IL-6andtransforminggrowthfactor-ßarerequiredforthedifferentiationofnaïveCD4+TcellstoTh17cells,whicharemaintainedinthepresenceofIL-23andIL-1ß.Conversely,IL-4andinterferon-?caninhibitthedevelopmentofTh17cells.Th17cellshavebeenimplicatedinthepathologyofmouseautoimmunediseasemodels.ExpressionofIL-17FandIL-17Ahasbeendetectedinactivatedhumanperipheralbloodlymphocytes.IthasbeenshownthatthecytokinesareexpressedinactivatedhumanCD4+Tcells.ExpressionofIL-17FandIL-17Ahasalsobeenobservedintissuesamplesfromvariousautoimmunediseasessuchasrheumatoidarthritis,multiplesclerosis,psoriasis,inflammatoryboweldisease,andasthma.ThecrystalstructureofIL-17Fhasbeensolvedandshowsthattheproteinformsadisulfide-linkeddimericglycoprotein.IL-17Aisalsoadisulfide-linkedhomodimericglycoprotein.TheIL-17Fhomodimerincludesaclassicalcysteineknotmotif,whichisfoundintheTGF-ß,bonemorphogeneticprotein,andnervegrowthfactorsuperfamilies.OnedifferenceinthecysteineknotmotifofIL-17Fcomparedwiththeotherknowncysteineknotproteinfamiliesisthatitonlyutilizesfourcysteinesinsteadoftheclassicalsixcysteinestoformtheknot.IL-17FandIL-17AhavebeenshowntoformBIOLOGicallyactiveIL-17F/IL-17Aheterodimers,inadditiontotheIL-17FandIL-17Ahomodimers.
