IntercellularAdhesionMolecule-2(ICAM-2)isa60kDasurfaceglycoproteinthatpromotesadhesionbetweenimmunecellsandthevascularendothelium.ICAM-2isamemberoftheIg-superfamilywithtwoimmunoglobulin-likeextracellulardomains.ICAM-2isbroadlydistributedonhematopoieticcells.Onrestinglymphocytes,theICAM-2expressionisseveral-foldhigherthanthatseenwithICAM-1,whilemonocytesexpressequivalentlevelsofICAM-1andICAM-2.IncontrasttoICAM-1,neutrophilsstainnegativeforICAM-2.JustlittleornoICAM-2stainingcanbedemonstratedonothercelllines,thesoleexceptionbeing(i)restingvascularendothelialcells,whichpossesshighlevelsofICAM-2,and(ii)smallclustersofcellsinlymphoidtissuegerminalcentres.ICAM-2expressionisunaffectedbyavarietyofinflammatorycytokines,whileICAM-1isupregulateduponstimulation.ThispointstowardtheimportanceofICAM-2intheunstimulatedrestingstate,beforeICAM-1expressionisincreased.TheintegrinLFA-1isthereceptorforICAM-2andICAM-1aswell.ThefunctionalcharacteristicsofICAM-2asLFA-1ligandcanbesummarizedasfollows:ICAM-2dominatesoverICAM-1onrestingendothelialcellsandseemstobeinvolvedintherecirculationofLFA-1positivelymphocytes,e.g.facilitatingT-memorycellrecirculation.AsrestingT-cellsexpresslittleornoICAM-1,ICAM-2mayalsobeimportantininitialT-celladhesionwithantigen-presentingcellsthatbearLFA-1.AlsothelysisofcertaintargetcellsappearstooccurinanICAM-1independentmanner,possIBLyregulatedbyICAM-2.
