Alzheimer’sdisease(AD)ischaracterizedbythepresenceofextracellularplaquesandintracellularneurofibrillarytangles(NFTs)inthebrain.Aggregationoftheamyloidβ-protein,especiallytheAβ42isoform,playsacriticalroleinthepathogenesisofAD.ShirasawaandIrieet.alhaveproposedatoxicconformerwithaturnatpositions22and23,aswellasanontoxicconformerwithaturnatpositions25and26,inAβ42aggregatesfromsystematicprolinescanningandsolid-stateNMRstudies.Thismonoclonalantibodynamed11A1wasdevelopedfortoxicAβ42,usingE22P-Aβ10-35,aminimummoietyforneurotoxicitycontainingtheturnatpositions22and23,forthegeneration.ImmunohistochemicalstudiesshowedthatnotonlyextracellularbutintracellularamyloidwasstainedinhumanADbrains,whichsuggestthat11A1coulddetecttoxicoligomersofAβwiththeturnatpositions22and23.
