Alzheimer’sdisease(AD)ischaracterizedbythepresenceofextracellularplaquesandintracellularneurofibrillarytangles(NFTs)inthebrain.Themajorcomponentoftheseplaquesisthebetaamyloidpeptide(Aβ),a40to43aminoacidpeptidecleavedfrombetaamyloidprecursorproteinbyβ-secretaseandaputativeγ-secretase.The‘longerforms’ofAβpeptide,Aβ42orAβ43,haveagreatertendencytoaggregatethanAβ40.IncreasedreleaseofthesepeptideshasbeenfoundtooccurinindividualswithgeneticmutationsoftheApoEalleles,andmightalsobecausedbyotherstillunknownmechanisms.ManyresearcherspostulatethattheincreasedreleaseofAβ42/Aβ43mayleadtotheabnormaldepositionofAβandtheassociatedneurotoxicityinthebrainsofaffectedindividuals.
