IL-4mediatesitsfunctionbybindingtoreceptorsexpressedontargetcells.ReceptorsexistonfreshlypreparedBandTlymphocytesandmacrophages,aswellasonvariouscelllinesincludinglymphoidcells,mastcelllines,avarietyofotherhematopoieticcelllines,fibroblastsandstromalcelllines.OnTandBcells,receptorsarepresentinlownumbers(appr.400),whicharereportedtobeup-regulatedbyIL-2andIL-4.IL-4isproducedbyaparticularsubsetofThelpercells,theTH2cells.ThesecellstendtomakeaspecificsetoflymphokinesincludingIL-4,IL-5,IL-6,IL-10,IL-13,IL-3andGM-CSFandfailtoproduceIL-2,IFNg,andlymphotoxin(TNFb).ApartfromTcells,ithasbeenshownthatmastcellscanproduceIL-4.IL-4exertsnumerouseffectsonvarioushematopoieticcelltypes.OnBcells,IL-4promotesimmunologicalclassswitchingtoIgEandIgG1isotypesandupregulatesMHCclassIIandCD23expression.Itcanpromotesurvival,growth,anddifferentiationofbothTandBlymphocytes,mastcells,andendothelialcells.Inaddition,IL-4caninhibittheproductionofTNF,IL-1,andIL-6bymacrophages.
