ProinsulinisaprecursorofinsulinthatissynthesizedinthebetacellsoftheisletofLangerhansinthepancreas.
Mostoftheproinsulinsynthesizedintheroughendoplasmicreticulumofbetacellsisstoredinthesecretorygranules.Proinsuliniscleavedbyprohormoneconvertingenzyme(PC)1/3atthejunctionoftheC-peptidewiththeB-chainofinsulin,subsequentlyitiscleavedbyPC2atthejunctionoftheC-peptidewiththeA-chainofinsulin,consequentlyitisdividedintoinsulinthatconsistsofa21amino-acidalpha-chainanda30amino-acidbeta-chainandc-peptidethatconsistsof33aminoacid.
Ithasbeenconsideredthattheconversionratefromproinsulintoinsulinintype2diabetespatientsdecreasesduetodroppingdownofPC1/3enzymaticactivitybyabnormalfunctionofpancreaticbetacells.Thus,intactproinsulinlevelinbloodortheratioofintactproinsulinandinsulin(P/Iratio)canbeconsideredtobecomeapotentialbioMarkerthatreflectsfunctionalchangeofthePC1/3activityatpancreaticbetacells.
Especially,IthasbeenknownthattheP/IratioisavaluablepancreaticbetacellevaluationmethodthatdoesnotrequirealoADIngtestasitcanbemeasuredusingbloodsamplesinfastedstate.
ThisELISAkitcanbeusedformeasuringintactproinsulin,notincludingcleavedintermediateproducts,inmouseandratsamples.
