Osteopontin(OPN)isknownasahighlyacidiccalcium-bindingglycosylatedphosphoproteinsecretedbymanycelltypes,includingosteoblasts,kidneytubulecells,macrophages,activatedTcells,andvascularsmoothmusclecells.Itsmolecularweightshavebeenreportedintherangeof66kDato44kDadependingonglycosylationandphosphorylation.OneimportantfeatureofOPNisthatitcontainsanArg-Gly-Asp(RGD)aminoacidsequence.Thismotifispresentinfibronectin,vitronectinandavarietyofotherextracellularproteinsthatbindmembersoftheintegrinfamilyofcellsurfacereceptorssuchasavß3.AnotherimportantpointisthepresenceofvariousmolecularformsofOPNinvivoduetodifferentialRNAsplicing,glycosylation,phosphorylation,sulfation,andsusceptibilitytoproteases.BothOPNandthrombinarelikelytobelocalizedtogetheratthesiteofinjury,inflammation,andangiogenesisandintumortissues.OsteopontinissusceptIBLetoproteolyticfragmentation,andthisprocessmayhavephysiologicimportance.ThekitcanspecificallyassayforRatOsteopontin(OPN),andcannotassaythrombin-cleavedRatOPNfragments.
