Cyclooxygenase(COX,alsocalledProstaglandinHSynthaseorPGHS)isabifunctionalenzymeexhibitingbothCOXandperoxidaseactivities.TheCOXcomponentconvertsarachidonicacidtoahydroperoxyendoperoxide(PGG2),andtheperoxidasecomponentreducestheendoperoxidetothecorrespondingalcohol(PGH2),theprecursorofPGs,thromboxanes,andprostacyclins.ItisnowwellestablishedthattherearetwodistinctisoformsofCOX,namelyCOX-1andCOX-2.Cayman’sCOXFluorescentActivityAssayprovidesaconvenientfluorescence-basedmethodfordetectingCOX-1orCOX-2activityinbothcrude(celllysates/tissuehomogenates)andpurifiedenzymepreparations.TheassayutilizestheperoxidasecomponentofCOXs.Inthisassay,thereactionbetweenPGG2andADHP(10-acetyl-3,7-dihydroxyphenoxazine)producesthehighlyfluorescentcompoundresorufinthatcanbeanalyzedusinganexcitationwavelengthof530-540nmandanemissionwavelengthof585-595nm.Thekitincludesisozyme-specificinhibitorsfordistinguishingCOX-2activityfromCOX-1activity.
