EndothelialLeukocyteAdhesionMolecule-1belongstotheselectinfamilyofadhesionmolecules.TogetherwithL-selectinandP-selectin,E-selectinmediatestheinitialinteractionsofleukocytesandplateletswithendothelialcells.Molecularstructure:Theextracellularpartofallselectinsconsistsofanaminoterminalc-typelectindomainwhichspecificallybindstocarbohydrateligands.ThisisfollowedbyanEGF-likedomain,and,inthecaseofE-selectin,by6shortconsensusrepeats.Thetransmembraneportionofthemoleculeisfollowedbyashortcytoplasmictail.Selectinsguidenon-activatedpolymorphonuclearcellstotheareasofinflammationincreatingfirst,loosecontactswiththeendotheliallayer.TogetherwithP-selectin,E-selectinisexpressedoncytokine-activatedendothelialcells,andcontributestotheadhesionofstillrestingleukocytestotheendothelium.Thisinitialbindingeventisanessentialprerequisitefortheactivationoftheimmunecellsviadifferentinflammatorymediators.IncontrasttoP-selectin,E-selectinismaximallyexpressed2-4hoursaftercellactivation.Withinthenext24-48hoursE-selectinisagaineliminatedfromthecytoplasmicmembranebysheddingintothecirculation.
