TumorNecrosisFactoralpha(TNF-alpha),alsoknownascachectin,isapolypeptidecytokineproducedbymonocytesandmacrophages.Itfunctionsasamultipotentmodulatorofimmuneresponseandfurtheractsasapotentpyrogen.TNF-alphacirculatesthroughoutthebodyrespondingtostimuli(infectiousagentsortissueinjury),activatingneutrophils,alteringthepropertiesofvascularendothelialcells,regulatingmetabolicactivitiesofothertissues,aswellasexhibitingtumoricidalactivitybyinducinglocalizedbloodclotting.TNF-alphaalsoinhibitslipoproteinlipaseactivityresultingincachexia,aphysicalwastingcondition.Duetoitsvariedactionsthroughouttheimmunesystem,TNF-alphamayplayaroleinthepathogenesisofmanydiseasestates.TNF-alphaproductionismediatedbytheactionoflymphokinesandendotoxinsonthemacrophage.PurifiedmonocytesproduceTNF-alphawithinfourhoursofstimulationbyrecombinantIL-2andthereissomeinvitroevidencetosuggestthatTNF-alphaisexpressedathighlevelsandwithprolongedkineticsinTcellsstimulatedbybothCD2andCD28.SecretionofTNF-alphaisenhancedbygammainterferon.TNFtheninducesorenhancesthespecificproductionofClassIMHCantigen,GM-CSF,andIL-1.Recentevidencehassuggestedanintracellularroleforthispeptide.
