Programmedcelldeathorapoptosisisthemostcommonformofeukaryoticcelldeathandisfoundduringtumorregressionandembryonicdevelopment.IntheprocessofselectionandeliminiationofautoreactiveBandTcellsapoptosisisanimportantprocessandthereforeaprerequisiteforthehomeostasisoftheimmunesystem.Apoptosisischaracterizedbychangesincellularmorphology(e.g.nuclearcondensation,membranebledding)andbiochemicallybyrapidinductiononDNAfragmentation.APO-1/Fas(CD95),amemberoftheTNF/NGFreceptorsuperfamily,isaglycosylated48kDsurfaceproteincontainingasingletransmembraneregion.APO-1isexpressedonavarietyofhumanBandTcelllines,onmanydifferenttumorcellsandonvariousnormalhumantissues.TriggeringofAPO-1byitsligandorbycertainanti-APO-1monoclonalantibodiesresultsinrapidinductionofprogrammedcelldeath,apoptosis,insusceptIBLecells.ThetissuedistributionofAPO-1andoftheAPO-1ligandsuggeststhattheAPO-1receptor/ligandsystemplaysanimportantroleinvariousaspectsofmammaliandevelopmentandespeciallyinthehomeostasisoftheimmunesystem.ElevatedsAPO-1levelshavebeenreportedinserafrompatientswithhigh-andlow-grademalignantB-andT-cellleukemiasandsystemiclupuserythematosus(Knippingetal.,submitted).sAPO-1maypreventcellsfromundergoingAPO-1ligandinducedapoptosis.Hence,secretionofsAPO-1mayprovideamechanismfortumorcellstoexcapeimmunosurveillanceandmaybeinvolvedinleukemogenesis.
