AnnexinIgG/IgMELISAisasolidphaseenzymeimmunoassayforthequantitativeandqualitativedetectionofIgGand/orIgMantibodiesagainstAnnexinVinhumanserum.TheassayemploysnativehumanAnnexinV.Theassayisatoolinthediagnosisoftheantiphospholipidsyndrome.AntibodiestargetingAnnexinVbelongtothefamilyofantiphospholipidantibodiesincludingforexampleanti-Cardiolipin,ß2-glycoproteinI(ß2-GPI)andProthrombinantibodiesandareassociatedwithprimaryandsecondaryantiphospholipidsyndrome(APS).Clinically,theAPSischaracterizedbyvenousandarterialthrombosisandseverepregnancycomplications.Anti-AnnexinVantibodieshavebeendetectedinpatientswithsystemicautoimmunediseases,especiallysystemiclupuserythematosus,buthavealsobeenassociatedtointrauterinefetalloss,recurrentabortionsandpreeclampsiainAPSpatients.AnnexinVisaCa2+-dependent,phospholipid-bindingproteinwithpotentanticoagulantactivitythatisabundantincellsexposedtoblood,suchasplatelets,trophoblasts,andendothelialcells.ThehighconcentrationofannexinVinplacentalsyncytiotrophoblastshasbeenproposedasamechanismtomaintainbloodfluidityonthesurfaceoftheplacentaandplacentalintergrity.ItthereforeunsuressupplyofthefoetuswithnutrientsandfetalviABIlity.ThrombosisandpregnancylossintheantiphospholipidsyndromemaybecausedbydisruptionofthisAnnexinVshieldbyantiphospholipidantibodiesleADIngtoanincreaseinthequantityofthrombogenicphospholipidsexposedtothecirculatingblood.Anti-AnnexinVautoantibodiesaresupposedtointerferewiththeanticoagulantfunctionofAnnexinVcausingplacentalthrombosisanddisruption.Consequently,thedetectionofanti-AnnexinantibodiesshouldbeincludedintheSEROlogicaldiagnosisofAPSespeciallyinfemalepatientsatriskforobstetriccomplications.
