IL-4mediatesitsfunctionbybindingtoreceptorsexpressedontargetcells.TheIL-4receptorsexhibitanaffinityofapproximately10-10M.ReceptorsexistonfreshlypreparedBandTlymphocytesandmacrophages,aswellasonvariouscelllinesincludinglymphoidcells,mastcelllines,avarietyofotherhematopoieticcelllines,fibroblastsandstromalcelllines.OnTandBcells,receptorsarepresentinlownumbers(appr.400),whicharereportedtobeupregulatedbyIL-2andIL-4.IL-4isproducedbyaparticularsubsetofThelpercells,theTH2cells.ThesecellstendtomakeaspecificsetoflymphokinesincludingIL-4,IL-5,IL-6,IL-10,IL-13andGM-CSFandfailtoproduceIL-2,IFN-gamma,andlymphotoxin(TNF-beta).ApartfromTcells,ithasbeenshownthatmastcellscanproduceIL-4.IL-4exertsnumerouseffectsonvarioushematopoieticcelltypes.OnBcells,IL-4promotesimmunologicalclassswitchingtoIgEandIgG1isotypesandupregulatesMHCclassIIandCD23expression.Itcanpromotesurvival,growth,anddifferentiationofbothTandBlymphocytes,mastcells,andendothelialcells.Inaddition,IL-4caninhibittheproductionofTNF,IL-1,andIL-6bymacrophages.
